Title: “A system-based investigation of the antibacterial mechanism of action of trans-translation inhibitors”
Abstract:
In bacteria, trans-translation is the primary ribosome rescue process. We employed proteome analysis of the response of Bacillus subtilis to gain insights into the physiological effects of trans-translation inhibition. The investigated small molecule inhibitors belong to different structural classes and target different molecules involved in trans-translation. In addition to targeting trans-translation, they synergize with metals for uptake and anti-Bacillus activity. Based on proteomics results, element analysis, in vitro studies and mutant analysis, we developed a model that could explain the mechanisms underlying the synergistic action.
